Los síntomas pueden indicar cálculos renales y el médico podrá determinar la presencia de éstos con sólo un examen físico.

Si se cuela la orina se pueden capturar cálculos del tracto urinario cuando son excretados, y su análisis muestra cistina. Un análisis de orina puede mostrar sedimento y cristales de cistina. Una recolección de orina en 24 horas muestra un exceso de cistina.

Una IRM o una TC abdominal , o un ultrasonido del abdomen pueden mostrar la presencia de cálculos en el tracto urinario. Así mismo, un PIV (pielograma intravenoso) puede revelar cálculos en el tracto urinario.
