Se puede presentar una hernia cerebral si se realiza en una persona con una masa en el cerebro como un tumor o un absceso, lo cual puede ocasionar daño cerebral o muerte. Por esta razón, una punción lumbar no se hace si otros exámenes muestran signos de tumor o absceso.

Se puede presentar una molestia temporal en las piernas si la aguja irrita la raíz de un nervio, pero esto desaparece cuando se retira la aguja.
